Metabolic complementation in bacterial communities: Necessary conditions and optimality

2016

Bacterial communities may display metabolic complementation, in which different members of the association partially contribute to the same biosynthetic pathway. In this way, the end product of the pathway is synthesized by the community as a whole. However, the emergence and the benefits of such complementation are poorly understood. Herein, we present a simple model to analyze the metabolic interactions among bacteria, including the host in the case of endosymbiotic bacteria. The model considers two cell populations, with both cell types encoding for the same linear biosynthetic pathway. We have found that, for metabolic complementation to emerge as an optimal strategy, both product inhib…

PET: Theoretical Background and Practical Aspects

2012

Positron emission tomography (PET) is a nuclear medicine imaging tool utilized for investigation of physiological processes in vivo. PET uses the decay characteristics of positron-emitting radionuclides which are produced in a cyclotron and then used to label compounds involved in physiological processes. Usually, the labeled compound—the tracer—is administered intravenously and distributed in the tissue. The radionuclide decays and the emitted photons are detected by the PET scanner. PET then offers the possibility to compute three-dimensional images of the biodistribution and kinetics of the regional radioactivity concentration. There are several options to analyze reconstructed PET image…

An experimental and kinetic modeling study on the oxidation of 1,3-dioxolane

2021

International audience; The modern catalytic or enzymatic advances allow the production of novel biofuel. Among them, 1,3dioxolane can be produced from formaldehyde and ethylene glycol, both can be obtained from biomass. In this study, the oxidation of 1,3-dioxolane is studied at stoichiometric conditions. The ignition delay times of 1,3-dioxolane/O 2 /inert mixtures were measured in a shock tube and in a rapid compression machine at pressures of 20 to 40 bar and temperatures ranging from 630 to 1300 K. The pressure profiles recorded in the rapid compression machine show a first stage of ignition enlightening the influence of the low temperature chemistry of combustion. Furthermore, mole fr…

Magnetic resonance pharmacokinetic imaging clusterization of hepatocellular carcinomas as a means to grade tumor aggressiveness.

2012

Hepatocellular carcinoma (HCC) management takes into account clinical and radiological findings, such as tumor stage, hepatic functional status and clinical symptoms. It is necessary to evaluate the number, size and location of the lesions. However, lesion aggressiveness is not considered in this therapeutic workflow, although the biology and the growth rate of the lesions have an important impact on survival. The aim of this work was to establish if the quantitative pharmacokinetic assessment of dynamic contrast-enhanced magnetic resonance images of HCC can separate lesions with different microvascular properties and biological evolution. Forty five patients with HCC and dynamic contrast-e…

The role of molecular oxygen in the formation of radiation-engineered multifunctional nanogels

2019

Abstract Nanogels are very promising biomedical nanodevices. The classic “radiation chemistry-based” approach to synthetize nanogels consists in the irradiation with pulsed electron beams of dilute, N2O-saturated, aqueous solutions of water-soluble polymers of the “crosslinking type”. Nanogels with controlled size and properties are produced in a single irradiation step with no recourse to initiators, organic solvents and surfactants. This paper combines experimental syntheses, performed with two e-beam irradiation setups and dose-ranges, starting from poly(N-vinyl pyrrolidone) solutions of various concentrations, both in N2O-saturated and air-saturated initial conditions, with the numerica…

Modeling of Zymomonas mobilis central metabolism for novel metabolic engineering strategies

2014

Mathematical modeling of metabolism is essential for rational metabolic engineering. The present work focuses on several types of modeling approach to quantitative understanding of central metabolic network and energetics in the bioethanol-producing bacterium Zymomonas mobilis. Combined use of Flux Balance, Elementary Flux Mode, and thermodynamic analysis of its central metabolism, together with dynamic modeling of the core catabolic pathways, can help to design novel substrate and product pathways by systematically analyzing the solution space for metabolic engineering, and yields insights into the function of metabolic network, hardly achievable without applying modeling tools.

Physiologically-Based Pharmacokinetic/Pharmacodynamic Model of MBQ-167 to Predict Tumor Growth Inhibition in Mice

2020

MBQ-167 is a dual inhibitor of the Rho GTPases Rac and Cdc42 that has shown promising results as an anti-cancer therapeutic at the preclinical stage. This drug has been tested in vitro and in vivo in metastatic breast cancer mouse models. The aim of this study is to develop a physiologically based pharmacokinetic/pharmacodynamic (PBPK-PD) model of MBQ-167 to predict tumor growth inhibition following intraperitoneal (IP) administration in mice bearing Triple Negative and HER2+ mammary tumors. PBPK and Simeoni tumor growth inhibition (TGI) models were developed using the Simcyp V19 Animal Simulator. Our developed PBPK framework adequately describes the time course of MBQ-167 in each of the mo…

An experimental and modelling study of the oxidation of 3-pentanol at high pressure

2018

International audience; High pressure oxidation of 3-pentanol is investigated in a jet-stirred reactor and in a shock tube. Experiments in the reactor were carried out at 10 atm, between 730 and 1180 K, for equivalence ratios of 0.35, 0.5, 1, 2, 4 and 1000 ppm fuel, at a constant residence time of 0.7 s. Reactant, product and intermediate species mole fractions were recorded using Fourier transform infrared spectroscopy (FTIR) and gas chromatography (GC). Ignition delay times were measured for 3-pentanol/O2 mixtures in argon in a shock tube at 20 and 40 bar, in a temperature range of 1000–1470 K and for equivalence ratios of 0.5, 1 and 2. The fuel did not show any low-temperature reactivity…

Three zone modeling of downdraft biomass gasification : equilibrium and finite kinetic approach

2013

Mathematical models and simulations are being practiced exceedingly in the field of research and development work. Simulations provide a less expensive means of evaluating the benefits and associated risk with applied field. Gasification is a complex mechanism, which incorporates thermochemical conversion of carbon based feedstock. Therefore, simulation of gasification provides a better comprehension of physical and chemical mechanism inside the gasifier than general conjecture and assist in optimizing the yield. The main objectives of present thesis work involve formulation of separate sub-model for pyrolysis and oxidation zone from published scientific references, and assembling it with p…
